Mono-acylated variant of a genetic fusion comprising the C-terminal folding scaffold of RTX block V (residues 1562–1681 of CyaA) and the CyaA residues 719–1294

Derived from adenylate cyclase toxin, the protein spans from residue 719 to 17006, harboring a deletion in the RTX domain (residues 1295-1561, inclusive). Recombinant protein, expressed in Escherichia coli, with a single acyl modification in position K272, corresponding to residue 983 of CyaA. A conservative substitution in residue K149 (K860 of CyaA) avoids the acylation by the dedicated acyl transferase CyaC. The monomers of the protein were obtained by molecular confinement in Sephadex 200 10/300. The sample was concentrated using centricon cutoff 10kDa.
