HlyA sequence with an additional polyhistidine-tagged eGFP (green fluorescent protein) fused to the N-terminus (amino acids 1-265 of the construct used for SAXS). The native HlyA sequence also contains and an engineered Flag tag inserted between amino acids 716-717 (UniProt numbering). HlyA is the substrate of the Hly T1SS complex and is transported unfolded, and then folds in the extracellular space. The fast folding GFP stalls the Hly T1SS complex because it cannot enter the channel in a folded state.
