SAXS data were collected on SARS-CoV-2 NSP15 in solution. The protein forms a hexamer with a theoretical molecular weight of approximately 234 kDa (6 × 39.0 kDa), consistent with the oligomeric state observed under our assay conditions. In the presence of the ADPRP (Mac1) macrodomain, the SAXS profile corresponds to a hexameric NSP15 bound to a single ADPRP macrodomain, giving an expected molecular weight of approximately 253 kDa (NSP15 hexamer ~234 kDa plus ADPRP ~19 kDa), which agrees well with the SAXS-derived molecular weight estimates.
