Synchrotron SAXS
data from solutions of
SEC-SAXS of Human microtubule-associated protein tau dGAE fragment (2N4R residues 297–391)
in
PBS, pH 7.4
were collected
on the
EMBL P12 beam line
at the PETRA III storage ring
(DESY; Hamburg, Germany)
using a Pilatus 6M detector
at a sample-detector distance of 3 m and
at a wavelength of λ = 0.123987 nm
(I(s) vs s, where s = 4πsinθ/λ, and 2θ is the scattering angle).
In-line size-exclusion chromatography (SEC) SAS was employed. The SEC parameters were as follows: A 40.00 μl sample
at 7.1 mg/ml was injected at a 0.70 ml/min flow rate
onto a GE Superdex 200 Increase 10/300 column
at 20°C.
2100 successive
1 second frames were collected.
The data were normalized to the intensity of the transmitted beam and radially averaged; the scattering of the solvent-blank was subtracted.
Funding: This research was funded by the EU NextGenerationEU through the Recovery and Resilience Plan for Slovakia under the project No. 09I03-03-V04-00623.
CAUTION: The SEC-SAXS chromatogram exhibits a pronounced, non-stationary baseline across the elution region. Because a unique stable background could not be defined, multiple scattering profiles obtained by subtraction using alternative background regions are provided in an additional ZIP file. These profiles represent alternative processing of the same SEC-SAXS measurement. The derived Rg, I(0), molecular mass, Dmax and P(r) should therefore be interpreted with caution.
