Synchrotron SAXS
data from solutions of
ABC transporter TM287/288 heterodimer in complex with nanobody Nb_TM#1
in
20 mM HEPES, 200 mM NaCl, 5 mM MgCl2, 20 μM DMM, 1 mM Mg2+ ATP, pH 7.6
were collected
on the
EMBL P12 beam line
at the PETRA III storage ring
(DESY; Hamburg, Germany)
using a Eiger 4M detector
at a sample-detector distance of 3 m and
at a wavelength of λ = 0.124 nm
(I(s) vs s, where s = 4πsinθ/λ, and 2θ is the scattering angle).
at 20°C.
100 successive
10 second frames were collected.
The data were normalized to the intensity of the transmitted beam and radially averaged; the scattering of the solvent-blank was subtracted.
Time-resolved experiments: final scattering curves collected 240 s after mixing. The remaining kinetic curves are provided as supplementary files. The discrepancy between the theoretical and experimental molecular weight arises from the presence of DDM detergent molecules bound to the TM287/288 heterodimer for solubility.
